Mulaney was always certain that he wanted to work in the entertainment industry. He became a member of “The Rugrats,” a children's sketch group in Chicago when he was seven years old. John's parents declined an audition for the part of Kevin in the movie “Home Alone” because of the club.

Mulaney went to middle school in Chicago at St. Clement School. Instead of writing reports, he and his best friend put on skits about what they had learned. When he was 14 years old, John played Wally Webb in the theatre performance of “Our Town.”

Mulaney attended St. Ignatius College Prep before enrolling at Georgetown University, where he studied religion and English. John joined the improv group at his university, where he met Nick Kroll and Mike Birbiglia. Later, he went on tour with Birbiglia performing stand-up comedy, which helped him develop his stage persona.

Mulaney relocated to New York City to pursue a career in comedy after receiving his degree from Georgetown in 2004. He was shortly given a position at Comedy Central as an office assistant. After working for the television network for a year, John suggested an idea for “I Love the '30s,” a parody of “I Love the '80s,” which he and fellow comic Nick Kroll had written. Mulaney abandoned his job to concentrate on his career after realising it was a “hindrance to being a comic.”

With stand-up specials including The Top Part (2009), New in Town (2012), The Comeback Kid (2015), and Kid Gorgeous (2018), for which he received a Primetime Emmy Award for Best Writing for a Variety Special, Mulaney is as well known for his work as a stand-up comedian. John Mulaney & the Sack Lunch Bunch, a children's musical comedy special on Netflix, is another famous work by Mulaney.

Mulaney wed Annamarie Tendler, a lampshade maker and cosmetics artist, in 2014 but unfortunately, they got divorced in 2021.

In 2015, Mulaney wrote on Instagram about the first night he and Tendler spent in their new home “It was our first night in our new home. We don't have any furniture, but we do have a leopard made of ceramic, so that's where we are. We're pretty sure it's not haunted, but if it is, we're ready for a fight. A lovely wife deserves a lovely home.” Their dog was also in the picture. Who will end up owning this house is still unknown. But the divorce settlement will take money away from Mulaney.
